Improved Dose-Response Analyses Using Direct Titration With Picoliter Dispense
Serial dilution is ubiquitous in preclinical discovery, used for dose-response analyses from assay development to lead optimization, ADMET and drug-drug interaction studies. This sequential process has numerous known problems such as carry-over, compounded errors and compound wastage among others.
However, with HP picoliter to microliter dispense technology, direct titrations without serial dilution are performed at your bench top. This is accomplished by non-contact dispensing of the proper number of picoliter droplets into each well to independently generate each required dose. Any dose can be placed in any well on the plate, eliminating the typical constraints of serial dilution.
This use of digital dispense leads to numerous performance advantages. Four major ones are highlighted here: improvement in replicate precision, reduction of the impact of plate edge effects, greater Hill slopes, and precision improvements due to finely spaced doses.
In addition, this technology simplifies does-response workflows by eliminating intermediate plates and replication. It is well-suited for titrations into both dry plates and assay-ready plates, and into either enzymatic or directly into pre-plated cellular assays.
